The A64 is closed both ways in Ryedale at Barton Hill near Castle Howard.
There’s been an accident between the Welburn turn off and Barnygate Lane.
Traffic is coping well, but drivers are being asked to avoid the area.
The closure has now been extended to Old Malton.
North Yorkshire Fire and Rescue Service say they were called to the scene at 3.39am.
A spokesperson said: “Crews from Malton responded to a report of a single vehicle road traffic collision in which the car collided with a tree.
“Crews assisted ambulance crews and made the vehicle safe and before providing lighting to assist police. The incident was left in the hands of the police.”
A North Yorkshire Police spokesman said: “Due to a serious road traffic collision a section of the A64 Malton Bypass, near to Welburn village, has been closed in both directions.
“This is the section where the dual carriageway turns into a single carriageway.
“Diversions are being put in place and we request that motorists please avoid travelling on this route. The road is likely to remain closed for some time.”

A riverside path in York has closed due to "structural damage".
TRIBUTES are being paid to war veteran Joseph Thomas who has died aged 99 and will have a memorial funeral service at York Minster tomorrow (Wednesday, January 22).
